Job Description:

The Laboratory Manager is responsible for lab management, lab operations, lab member training, lab safety and lab maintenance. Further, this position is responsible for coordinating translational science collaborations and to develop and analyze patient and donor derived tissue samples and cell cultures via biochemical, immunological, and functional analyses. Finally, this position is responsible for managing databases for Lab Equipment, DNA stocks, RNA stocks and human/mouse cell stocks in support of a large cancer biology research lab in the Department of Pathology and Laboratory Medicine and the Legorreta Cancer Center at Brown University.

Research in Dr. Sobol’s lab focuses on mechanisms of DNA repair, PARP and NAD+ metabolism in human cells and the convergent role of these pathways in response to environmental genotoxins and chemotherapy. A major goal in the lab is to use biochemical, genetic, and imaging modalities to study DNA repair protein complexes that respond to DNA damage induced by environmental genotoxins and chemotherapy and how this pathway affects the regulation of cellular metabolism via ADP-ribosylation signaling and alterations in NAD+ metabolism.  Recent efforts have focused on the role of DNA repair pathways and the regulation of PARP1/PARP2 activation in response to replication stress, identification of novel targets to overcome PARP-inhibitor resistance and the development of novel biological tools and assays to evaluate genotoxic stress and PARP-activation that may be used for the discovery of novel compounds that may be used in the treatment of cancer and evaluating genotoxin response.

Techniques used in the lab include the genetic manipulation of cells in culture, genetic and epigenetic analysis of tumor cells, development and phenotypic characterization of transgenic cells, development and characterization of transgenic mice, RNA interference, CRISPR/cas9 gene editing, lentivirus production and viral transduction of mouse and human cells, molecular cloning of mouse and human cDNAs, DNA damage and repair assays, cell culture, cell cytotoxicity assays and PCR.

This is a fixed term position with a current end date of one year from hire, which may be extended based upon available funding.

Education and Experience

  • Bachelor’s degree in Molecular Biology, Biology, or related field required.
  • Master’s degree preferred.
  • At least 5-7 years’ experience working in organized research, or equivalent combination of education and experience required.
  • 2 years’ experience bench research preferred.
  • Experience in culturing human primary cells and development of primary cell lines from human tumors is preferred.
  • Experience in immunoblotting, immunohistochemistry, and qRT-PCR preferred.
